The Hollywood couple who wed in July 2010 will become parents for a second time later this year according to the New York Post.
Sources close to the actress have allegedly revealed her big news to the publication, adding that she is "around three months pregnant," but her spokesperson is yet to comment on the reports.
Penelope and Javier already have a two-year-old son called Leo who was born in January 2011.
The 38-year-old Spanish beauty has previously spoken of her love for motherhood in an interview with US Vogue where she called it a "revolutionary experience".
She said: "From the first second, you feel so much love. That's the best way I can describe it. It transforms you completely, in a second."
